Provigil is available in the market under multiple names, two of these are Vigicer and Aletec. Actually, Provigil is an FDA approved psychostimulant used to treat excessive sleepiness and sleep disorders. Besides that, Provigil is involved in:

  • Memory improvement
  • Mood enhancement
  • Increasing alertness
  • Cognitive enhancement

How it Acts

Like racetams, Provigil also works by increasing neurotransmitter activity in the brain. It achieves this by releasing histamines and increasing dopamine. Both chemicals are the ones that our body uses for rapid response stimulation. That means, with Provigil, your body is able to achieve heightened mental alertness and sharpness.

How Provigil Acts

Why do we need a drug that keeps us up anyway? We need it because without Provigil, lack of sleep leaves us unproductive and unable to perform to the best of our capabilities. With our lifestyles so fast paced and the job demands that just keep rising, we have all known sleep deprivation in one form or another. It may be an all-nighter required to do preparation for an exam. It could also be staying up the whole night to complete a report due at work the next day.

Research tells us that when compared, lack of sleep may be even worse than alcohol consumption! A study compared people who drove with a blood alcohol level of 0.05% fared better than those who drove after having stayed up for 17 hours or more! If these reasons are insufficient, here are some advantages that come with the use of Provigil:

  • One of the main advantages is that it makes you feel fresher when compared to amphetamines.
  • Secondly, Provigil allows you to stay alert, as well as, awake for more than 40 hours.
  • Yet another advantage of using Provigil is that unlike caffeine, you do not crash when the drug wears off. All you have to do is catch up on some sleep and you are good to go! No more jitters that are so common with dextroamphetamine, caffeine, and cocaine.

Safety & Side Effects

Provigil tablets are available as either 100 mg or 200 mg. While the dose that works for you will depend on your own neurochemistry, 100 mg is said to be sufficient. With this dose, most users who use it as a cognitive enhancer, report they function well throughout the day. Some people will take 200 mg only on intense days when they need that extra boost. However, Provigil is expensive and small doses like 100 mg should do the trick, as well as, be light on your wallet. Provigil has a life of about 12 hours. That means if taken at 7 in the morning, you can depend on its effects to continue until around 7 in the evening.

Drug Interactions

CYP3A4/5 Substrates

Drugs that are substrates for CYP3A4/5 include:

  • Cyclosporine
  • Midazolam
  • Steroidal contraceptives
  • Triazolam

The doses of these drugs will need to be readjusted since Provigil causes induction of metabolic enzymes. Moreover, Provigil makes the contraceptives mentioned above less effective when co-administered, even a month after that. It is recommended that alternative methods of contraception are used with Provigil.

CYP2C19 Substrates

Drugs that are substrates for CYP2C19 include the following:

  • Clomipramine
  • Diazepam
  • Phenytoin
  • Propranolol
  • Omeprazole

The doses of these drugs will need to be readjusted if they are being taken with Provigil. This is because the latter inhibits metabolic enzymes.

If co-administered with Warfarin, prothrombin times need to be monitored more frequently.

Monoamine Oxidase (MAO) Inhibitors

Be careful if you are taking Provigil and MAO inhibitors at the same time.
